Probably, yes.  If you mean one card dedicated to one monitor and the other to the other monitor, I'm not too sure.  Depends on the multi-GPU tech being used (SLI/Nvidia or CFX/AMD).

For your regular desktop and etc., it will work yes.  If you're talking about games... that may be complicated.

games will launch on your primary monitor on the primary GPU (designated to be monitor 1)
while you can drag other things onto the other window

For regular desktop.  I don't really want a two montior gaming thing because then every time I aim down the sight I have a black bar in the way. :P

For regular desktop it should definitely work.  I have two monitors plugged into my single GTX460 so it should work.  Try it out when you can.

I never do dual monitor gaming, mainly because one is 1440x900 and the secondary is 1024x768 lol.

I have an integrated nVidia 6150SE and Radeon HD 4550, Acer monitor is plugged into 4550 via DVI and Dell monitor plugged into nVidia via VGA.  HOWEVER the problem right now is that Windows 7 will not recognize the integrated.
