Correct me if I'm wrong, but didn't Twilight hatch the egg and has cared for Spike his entire life? That may not be a biological mother, but she would still be the closest thing to one he's ever had.

Twilight did hatch the egg and it's safe to assume she took care of Spike all of her life, but it seems to me that most of the time she thinks of him as more of an assistant than a family member.
