Using two short (3-5 car) trains to supply a station with a large (11-15 car) train, and then running the large train when it's fully loaded to a distant drop-off point, can make a hell of a profit.

Use path signals along your tracks to allow multiple trains on them at once, and remember to click the signal a second/third time to make it point the right way.
Use small supply lines to transfer goods to a larger station, then have large trains full load at it and head off to a pretty distant drop-off point.

Well I have the signals figured out but I guess I have too huge of trains at the supply points.

Aha! I have now beat Wedge in city population with my city, Duckville, consisting of 40,000 people, to his 35,000!
