No it doesn't. Sure NV uses the same engine, along with many of the same weapons, models and animations, but it can be taken into an entirely different context from F3. You could skip playing F3 altogether and still get NV's story completely.

So I was walking south from Canterbury Commons following the caravan, when I spotted a tent of sorts a quarter mile away. Going into sneak mode to get a closer look, a red [DANGER] was flashing at the top of the screen. Rushing into battle with Fisto upon my arm, I found a steel structure surrounded by turrets. An enemy was firing on me with a plasma pistol. Targeting him, I discovered he was an Enclave officer. When I killed this assailant, I focused on the seemingly innumerable turrets firing on me. I circled the structure clockwise, destroying every turret. When the last of them had finally popped its irate clogs, I examined the body of the late officer. I stole his outfit, having noted the fantastic cap upon his head before he died. I entered the tent to find several ammunition boxes and a computer terminal. After raiding the ammunition boxes, I took a look at the terminal:

Interesting, I thought as I closed the terminal. I got a pretty nice hat and outfit as well.

Almost at page 100. Yay.

Just beat Fallout 3. Thought I could leave the rotunda after coming in to activate the purifier. So I saved right outside the airlock.
Beat the game, reloaded, realized I couldn't leave. I just threw away an awesome level 18 character. :c
